Cardiff held firm for over an hour, with goalkeeper Ethan Horvath making seven key saves, but Villa finally broke through when Rashford and Asensio combined for the opener.
Asensio slotted in Rashford's cut-back in the 68th minute to break the deadlock, then doubled his tally in the 80th minute, securing Unai Emery's side a spot in the next round.
Aston Villa now awaits Sunday evening's draw, set to follow Manchester United's clash with Fulham at Old Trafford, to learn their quarter-final opponent.
Villa controlled the match and probably should have taken the lead well before their opening goal, as Ollie Watkins and Rashford, still searching for his first goal in claret and blue, both missed chances from close range before halftime.
